The fastest option on paper is train to Nashik Road plus a short taxi hop — here's the route, timings, and whether one day is realistic.
The standard route is NH60, running north through Chakan, Narayangaon, Sangamner and Sinnar before it meets Nashik city. From Nashik you pick up the Nashik-Trimbak road for the final stretch.

Expect 4-5 toll plazas on the way, adding up to roughly ₹200-300 one way for a car, less with FASTag. At Nashik you have a choice: drive through the city itself, or take the Sinnar-Trimbak bypass that skips the worst of the traffic lights if you're behind the wheel yourself.
Once you're close, every vehicle — including private taxis — is parked at the temple junction, not right outside the sanctum.
Driving isn't the only way in. MSRTC runs a handful of direct Pune-Trimbakeshwar services from the Shivajinagar (Wakdewadi) stand, mixing ordinary and semi-luxury Shivshahi buses through the day. The ride takes about 6 hours — check msrtcors.com close to your travel date since timings shift.
If you'd rather not sit on a bus that long, train to Nashik Road is the quicker combination. Several trains a day cover the route in roughly 3 hrs 45 min to 4 hrs, after which you're looking at a taxi or shared jeep for the last 28-30 km.

There's no direct train into Trimbakeshwar itself — Nashik Road is as close as the railway gets.
It's doable, but only with an early start. A private taxi leaving Pune by 5:00-5:30 am gets you to Trimbakeshwar by around 10:00-10:30 am, ahead of the worst of the darshan queue.

10:30 am-1:00 pm — darshan and a walk around Kushavarta Kund

On the ST bus this plan falls apart — a 6-hour one-way ride means 12 hours just in transit, leaving barely any time at the temple. The bus works far better as an overnight-stay trip than a same-day one.

Elders or a family group — private taxi. No transfers, no standing in a bus queue, and the driver waits at the temple.
Solo or budget travellers — the direct ST bus is the cheapest way in, if you can live with a fixed departure and a long day.
Tight on time from central Pune — train to Nashik Road is the fastest single leg, but only if the connecting taxi timing works for you.
Planning to add Saptashrungi or Brahmagiri — you'll want your own vehicle either way; public transport between these stops is thin.
Avoid the ST bus if you're doing this as a same-day trip — the fixed 6-hour timings rarely leave enough daylight for an unhurried darshan.
Most Yatris doing this as a day trip skip an overnight stay altogether. If you're combining Trimbakeshwar with Saptashrungi, Brahmagiri or Nashik's own temples, staying over makes more sense than two long drives on consecutive days.
